Cover Letter A cover letter is required. It must briefly explain what was previously known, the conceptual advance provided by the findings, and the significance of the findings to a broad readership.
Abstract An abstract is required. 250 words or fewer. Unstructured.
Manuscript The main text must include abstract, introduction, material and methods or subjects and methods or methods, results, and discussion. A Declaration of Interests section, a Web Resources section if applicable, a Data and Code Availability statement, and a supplemental information description titled "Supplemental Information" must be included when relevant.
References Only published articles may be included in the reference list.
Figures & Tables Each figure must be uploaded individually as a separate file, tables must be included after the figure titles and legends in the manuscript, and supplemental information may be uploaded as a single PDF except for very large tables of data or Excel files. Each figure must be numbered consecutively with whole numbers, figures may not exceed one page, figure titles must not contain parenthetical information, reference citations, or footnotes, and tables must not be saved as figures.

References No more than 30 references. References must include only published articles.
Figures & Tables No more than four display items. Figures and tables are uploaded separately from the manuscript, with figures submitted individually as separate files and tables included in the manuscript. Each figure may contain no more than four subpanels, and a provisional Mendelian gene discovery submission must include a table mapping phenotypic findings to HPO terms or IDs as one of the four display items.
